| 体外活性 | P450催化的Dibenzylfluorescein代谢及氢氧化钠影响的实验方法[3]如下:Dibenzylfluorescein在P450的作用下脱烷基形成荧光素苄基酯,氢氧化钠的加入能促进其水解为荧光素;而2M氢氧化钠可直接将Dibenzylfluorescein分解为荧光素苄基醚。实验条件包括:1) CYP2C19酶活性及抑制反应,将0.1M Tris-HCl (pH 7.4)、10 μM Dibenzylfluorescein、15 pmol CYP2C19和50 μL NADPH再生系统混合,形成150 μL反应体系。NADPH再生系统包含1.13 mM NADP、12.5 mM异柠檬酸、56.33 mM KCl、187.5 mM Tris-HCl (pH 7.4)、12.5 mM MgCl2、0.0125 mM MnCl2和0.075 U/ml异柠檬酸脱氢酶。2)样品在37°C下孵育30至60分钟,然后迅速冷却至4°C以终止反应,并经离心后取上清液进行LC-MS分析。3)对照分析在无氢氧化钠和加入2M氢氧化钠的条件下,使用纯Dibenzylfluorescein、荧光素苄酯、荧光素苄醚及荧光素(浓度均为10 μM)作为标准品。 |
